Can Adding PepGen to Bio-Oss Enhance Bone Growth?

I have heard several times at implant seminars that adding PepGen P-15 [Dentsply] to Bio-Oss [Osteohealth] can enhance the amount of bone growth. PepGen P-15 is supposed to be a very potent osteogenic agent, producing bone fill in infrabony defects in a very short time. It is also putty-like in consistency and easy to mold into place to fill defects. I have also read it somewhere in the literature. Has anyone tried it? Can anyone please tell me if there are any publications/journal articles that describe this use? I am also concerned that I cannot find a specific reference that recommends PepGen P-15 for use with implants – for instance to fill in a space around the implant as in an immediate insertion after extraction.

Pepgen is a fantastic product, even as a standalone. Putting the putty immediately into the osteotomy might impede the implant, however, Pepgen Flow works nicely in that case.

P-15 is a GTR material and is good arond teeth, and yes I would agree that it would help growth because it is an osteoinductive material. Now Bio-Oss is a Xenograft material that has a very hard time fully turning over into 100% live living bone. I would suggest a great course to learn more about bone and what types to use in specific applications. Dr. Don Callan puts on a wonderful course, which is rich in demonstrating scientific results over opinions. It is purely literature based and a great course to learn more about all the types of options you have out in the marketplace.
Look for bone types that are low on mineral content...these materials are osteoinductive...more BMP action.
Again I highly suggest Dr. Callan's course.

Pepgen p-15 is an excellent product, but we do not advocate its use by itself. we recommend mixing it with pepgen flow, in what we call a marshmallow technique. the flow expands and fills the socket completely. a description of this technique can be found at http://www.alvelogro.com/Publications/TB%20Marshmallow.pdf

This is the first time that I have heard that P-15 is osteoinductive. What type of assay is conducted to determine its inductivity?

I concur with Ray. It would be exciting to see data suggesting the "osteoinductive" properties of this product. It has been shown that the the P-15 protein binding MAY allow for better proliferation of osteoblasts...as evidenced by increase ALP levels but I don't read that as being inductive but.... Remeber that you still need the synergistic osteoclastic activity to produce the desired long-term effect of living bone. Hopefully someone will link to some valid articles.

Im sorry but it amazes me that there are dentist out there that use a product and dont have the first clue at what they are putting in the patient.
first off - PEPGEN 15 is essentially bio-oss with P-15 peptide attached to it. So the question that was posed was silly in the sense that your asking if mixing Pepgen 15 mixed with Bios-oss would make it better. Give me a break. KNOW YOUR PRODUCTS. PEPGEN 15 IS BOVINE BONE.
Next the answers that people give truly show that you dont read the research. Most of the Guru's out there stopped using Pepgen-15 long ago. READ THE RESEARCH. Bottom line its not Osteoinductive, it will not result in regeneration of Bone, cementum OR PDL but usually gets either fibous encapsulated or verly little bone forms around it. The other issue is it either never resorbs or takes forever to resorb.
The concept of PEPGEN 15 was sound but never worked. For educational purposes PepGen15 a synthetic biomimetic of the 15 amino acid sequence of Type-I collagen, which is uniquely involved in the binding of cells, particularly fibroblasts and osteoblasts4 and that is attached to bovine bone.
Read the research before praise a product. Most importantly read the product insert before you use it on a patient. Remember you have to do imformed consent prior to doing a bone graft and you need to inform the patient as to what you are using on them. Dont you think it would be nice if the Dentist actually knew that Pepgen15 is bovine bone.

I totally agree with DRK.P-15 is a bovine material.It is basically Osteograf-N plus the 15 amino acid sequence which is responsible for the cell binding to the collagen.It is necessary to understand that in order for this material to be effective it needs cells in the process of bone formation.
According to my long term experience both with the P-15 and the P-15 putty it is osteoconductive .

this is a question I posted up long time ago but had been modified by someone to the extend that I didn;t even realise it was a question I had asked. Bio-Oss and P15 are both bovine material. None of them is osteoinductive. I once read in the literature that adding both together would give you more bone formation (about 2 fold increase), compare with using either one alone. This was not because of its "osteoinductive" property of the P15, but because mixing large particle size (bio-oss) and small particle size (pepgen-15) would result in a mass of material that allow better ingrowth of blood vessels that lead to better bone formation. Pepgen15 when used alone the particle size was too small and may impeded blood vessels ingrowth. Has anyone read this article? or something alone the line?

Lot of scientists (Sottosanti, Walsh, Anson, ETC.) have recently shown that adding calcium Sulfate to allograft based bone grafts improves not only their handling characteristics, but also their performance significantly. So you might consider adding calcium sulfate to Bio-Oss for better performance. Recently a calcium sulfate based bone graft called DentoGen was featured on this forum.

P-15 is simply Osteograft-N with the 15 attached to to it. There is no good clinical data or any histology that I know of. Bio-Oss on the other hand has 20 years of successful cases world wide. The two products may both be bovine bone but are processed different. Bio-Oss looks like human bone under the SEM. I tried P-15 years ago when it first came out. I was very unhappy when I went back in to find mush and poor quality bone.
